ABSTRACT

Six dimensional syndrome differentiation theory, put forward by professor GU Xiao-hong at Beijing University of Chinese Medicine based on her theoretical teaching and clinical experience, emphasizes that the syndrome differentiation should be carried out from six dimensions including etiology, disease location, disease stage, disease condition, pathology, and disease nature, which is conducive to clinical thinking training and formation of traditional Chinese medicine (TCM). The differentiation and treatment of Baihutang syndrome frequently seen in cold damage and warm disease still need to be explored. Guided by the six dimensional syndrome differentiation theory coupled with diverse viewpoints of cold damage and warm disease schools, this paper summarized and reinterpreted the understandings and thoughts of GU Xiao-hong and YU He, warm disease specialists of two generations. Considering the lung-stomach dysfunction caused by the internal invasion of exogenous pathogens, Baihutang syndrome was staged into Qi aspect. In this stage, exuberant pathogens and sufficient healthy Qi allowed the prevailing of internal heat and the consumption of body fluid, manifested as high fever, profuse sweating, thirst, and the pulse corresponding to interior excess and heat syndrome. This paper also pointed out that the Baihutang syndrome involved both lung and stomach, and the adoption of Baihutang contributed to preventing tu from restricting shui in the case of extreme excess of Yang brightness and protecting the kidney Yin. As revealed by the dynamic analysis of prognosis of Baihutang syndrome based on the six dimensional syndrome differentiation theory, even though the Baihutang syndrome could be present in both cold damage and warm disease, the specific disease stage, transmission and change, condition, prognosis, pathology, and medication differed. On this basis, a series of prescriptions have been modified from Baihutang, which has expanded the application scope of Baihutang and enriched its research value, thus better promoting its clinical application.

ABSTRACT

OBJECTIVE@#To study the developmental and morphological characteristics of the mandible in patients with impacted mandibular second molar and to predict the possible trend of mandibular development via three-dimensional (3D) measurement and analysis.@*METHODS@#A total of 88 cases of impacted group and 88 cases of control group were screened out. 3D measurements were performed by using Mimics software. A total of 23 landmark points and 17 measurements were determined. The measurements were analyzed by t-test.@*RESULTS@#The mandible length, the space between the first molars, the space between mandibular angles, and the width between the first molars in the impacted group were lower than those in the control group (P<0.05). Moreover, the value of the submandibular angle was higher than that in the control group (P<0.05).@*CONCLUSIONS@#The impacted mandible of patients with mandibular second molar showed lack of sagittal and width development, and the impacted mandibular second molar was a manifestation of its degeneration.

ABSTRACT

Objective: To investigate the effect of hesperidin on apoptosis of gastric cancer AGS cells and its related molecular mechanisms. Methods: MTT assay was used for the killing effect of hesperidin on human gastric cancer AGS cells; Annexin V-FITC/PI double staining and flow cytometry was used to detect the apoptosis induced by hesperidin on AGS cells, the level of reactive oxygen species, and the addition of NAC Post-apoptosis changes; Western blotting was used to detect the expression of apoptosis-related proteins and signaling pathway-related proteins. Results: MTT assay showed that hesperidin had a good inhibiting effect on AGS cells. After treated with hesperidin, AGS cells showed apoptosis such as nuclear condensation and cell shrinkage. Annexin V-FITC/PI double staining and flow cytometry showed that hesperidin can induce mitochondrial dependent apoptosis of AGS cells and increase the level of intracellular reactive oxygen species. After pretreatment of NAC, hesperidin induced apoptosis inhibition. The results of Western blotting showed that the expression of p-JNK, p-p38, Bad, cleaved Caspase-3, and cleaved PARP increased, and the expression of anti-apoptotic proteins p-ERK and Bcl-2 decreased, which indicated that hesperidin activated the MAPK signaling pathway and mitochondria-dependent apoptosis in AGS cells. Conclusion: Hesperidin has a good killing effect on human gastric cancer AGS cells, and induces mitochondria-dependent apoptosis in AGS cells by increasing the level of reactive oxygen species in AGS cells and regulating MAPK signaling pathway.

ABSTRACT

Multidrug resistance (MDR) is one of the major obstacles in cancer chemotherapy. Our previous study has shown that icariin could reverse MDR in MG-63 doxorubicin-resistant (MG-63/DOX) cells. It is reported that icariin is usually metabolized to icariside II and icaritin. Herein, we investigated the effects of icariin, icariside II, and icaritin (ICT) on reversing MDR in MG-63/DOX cells. Among these compounds, ICT exhibited strongest effect and showed no obvious cytotoxicity effect on both MG-63 and MG-63/DOX cells ranging from 1 to 10 μmol·L. Furthermore, ICT increased accumulation of rhodamine 123 and 6-carboxyfluorescein diacetate and enhanced DOX-induced apoptosis in MG-63/DOX cells in a dose-dependent manner. Further studies demonstrated that ICT decreased the mRNA and protein levels of multidrug resistance protein 1 (MDR1) and multidrug resistance-associated protein 1 (MRP1). We also verified that blockade of STAT3 phosphorylation was involved in the reversal effect of multidrug resistance in MG-63/DOX cells. Taken together, these results indicated that ICT may be a potential candidate in chemotherapy for osteosarcoma.

ABSTRACT

The number of adults seeking orthodontic treatment is increasing. Shortening the course of orthodontic treatment has become a research hotspot in recent years. Methods of accelerating tooth movement have been reported. Theoretical and clinical studies have confirmed that periodontal corticotomy is effective in accelerating tooth movement and increasing osteogenic potential. Many surgical techniques of periodontal corticotomy have been developed. This review summarizes the history of periodontal corticotomy and the development of the surgical techniques in recent years, thereby providing references for the clinical application of corticotomy.

ABSTRACT

BACKGROUND: Cell-free stem cell therapy has been an issue of concern, but there is no conclusion on how to extract high-quality exosomes. OBJECTIVE: To extract exosomes from human umbilical cord mesenchymal stem cells by using three different methods, and then to screen the optimal method. METHODS: Exosomes were extracted from human umbilical cord mesenchymal stem cells by using the Total Exosome Isolation test kit, Exo Quick test kit and differential ultracentrifugation method, respectively. Then, transmission electron microscopy was used for morphological observations, BCA was utilized to quantify the protein, and western blot assay was applied to detect surface markers CD9, CD81 and CD63. RESULTS AND CONCLUSION: Extraction of exosomes was completed by all the three methods, and round or oval membranous vesicles were observed under the transmission electron microscope. The protein content and purity of exosomes was highest in the differential ultracentrifugation group, followed by the Exobiology Quick kit group, and lowest in the Total Exosome Isolation kit group, and there were significant differences among the three groups (P < 0.05). Under the same protein concentration, surface specific markers, CD81, CD63 and CD9, were expressed highest in the differential ultracentrifugation group, followed by the Exobiology Quick kit group, and lowest in the Total Exosome Isolation kit group. The operating time was significantly lower in the Exobiology Quick kit group compared with the other two groups (P < 0.05). To conclude, despite a longer operating time, the differential ultracentrifugation method is a rational method to extract enough exosomes with relative high purity.

ABSTRACT

Objective To explore the influence of long -term smoking on the level of blood inflammatory cytokines and the degree of atherosclerosis in healthy middle -aged men.Methods A total of 84 middle -aged men who accepted the health examination were divided into smoking group and non -smoking group.The risk factors of atherosclerosis,such as age,body mass index (BMI),systolic blood pressure (SBP),diastolic blood pressure (DBP),fasting blood glucose (GLU),total cholesterol (TC),low density lipoprotein (LDL),high density lipoprotein (HDL)and triglyceride (TG) were recorded.The white blood cell (WBC)level and neutrophil cell (NC)level were also recorded.The interleukin -6 (IL -6)concentration was assayed by using enzyme -linked immunosorbent double antibody method and the high -sensitivity C reactive protein (Hs -CRP)concentration was assayed by using immune turbidimetry method.The carotid artery intima -media thickness (IMT)and plaques were measured by B -mode ultrasonography.Results There were no significant differences in the risk factors of atherosclerosis between the two groups.But there were statistically significant differences between smoking group and non -smoking group in IL -6 (1 .46 ±0.27 vs.1 .21 ±0.24),Hs -CRP (6.57 ±2.47 vs.3.1 4 ±2.07),WBC (5.78 ±0.79 vs.5.25 ±0.64)and NC (3.40 ±0.45 vs.2.87 ±0.42)(P <0.05). The carotid artery IMT in smoking group was significantly thicker than that of the non -smoking group (1 .0 ±0.1 8 vs. 0.82 ±0.1 6,P <0.01 ).The detection rate of plaque and carotid stenosis in smoking group was significantly higher than that of the non -smoking group (P <0.01 ).Conclusion Long -term smoking not only leads to increased inflammatory cytokines,but also aggravates the degree of atherosclerosis in healthy middle -aged men.Long -term smoking healthy middle -aged men are potential in patients with cardiovascular disease.

ABSTRACT

<p><b>OBJECTIVE</b>To study the infection status of Leptospira in rodents on Heixiazi island Heilongjiang province in 2011.</p><p><b>METHODS</b>A total of 356 rodents were captured by night trap on the Heixiazi island from April to October 2011. The kidney tissue samples were collected by asepsis operation and the genomic DNA were extracted from them. Leptospira strains were confirmed by polymerase chain reaction(PCR) amplification of the 482 bp 23 S rDNA gene. Fifteen PCR products selected by the month were purified and sequenced by the methods of Sanger dideoxy, the sequences then compared with other Leptospira strains in Genebank, and phylogenetic analyses were drafted by software Mega 4.0.</p><p><b>RESULTS</b>Among 356 rodents, the dominant species were Clethrionomys rutilus (39.3%, 140/356) and Apodemus agrarius (36.0%, 128/356). The infection rate of Leptospira was 11.0%, with 39 rodent samples detected positive. All the rodent species were infected except for Rattus norvegicus. The infection rate was 9.4% (12/128) in Apodemus agrarius, 12.9%(18/140) in Clethrionomys rutilus, 10.8%(7/65) in Microtus fortis Buchner. No significant difference was found between the infection rate and the species of rodents by chi square test(χ(2) = 1.92, P > 0.05). Among months, the infection rate was 5.6% (4/72) in May, 8.8% (5/57) in June, 12.8% (5/39) in July, 9.8% (5/51) in August, 33.3% (11/33) in September, 22.5% (9/40) in October,but no infection in April. There was significant difference in infection in different months (χ(2) = 32.92, P < 0.05). All the Leptospira in rodents on the Heixiazi island were in the same phylogenetic branch with a high similarity of 97.1%-99.6%, close with the Australia strain U90865 by the similarity above 96.3%.</p><p><b>CONCLUSION</b>Leptospira is probably prevalent in rodents on the Heixiazi island, and the phylogene of the strains were similar. The infection rate in rodents was significantly different in months but not in hosts.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the dymamic accumulation of triterpenic acids production in submerged cultivation mycelium of Poria cocos.</p><p><b>METHOD</b>Liquid culture method of P. cocos was established and RP-HPLC was applied to determine the contents of three main triterpenic acids dehydrotumulosic acid (DTA), 3-epi-dehydrotumulosic acid (eDTA) and polyporenic acid C (PAC) in submerged cultivation mycelium P. cocos at different culture stages and the contents were compared with cultivated P. cocos. The HPLC method is as follows, column: Plastisil ODS (4.6 mm x 250 mm, 5 microm); mobile phase: ACN/0.5% phosphate (80:20); flow rate: 1.0 mL . min-1; detective wavelength: 242 nm.</p><p><b>RESULT</b>The maximum biomass occurred at the 8th d after inoluctation, however, the contents and yield of three compounds increased till the 17th day. The contents of three compounds were 1. 2% (DTA), 0. 42% (eDTA) and 1.0% (PAC) at the 17th day after inoculation, which were significantly higher than that in cultivated material [0.2% (DTA), 0. 12(eDTA) and 0. 16% (PAC) ]. Furthermore, a correlation analysis between the content ratios of three independent compounds was carried out. The results showed that DTA negatively correlated with eDTA and PAC, with R2 of 0. 857 6 and 0. 971 7, respectively, which suggested the role of DTA as an important intermediate in the biosynthesis of triterpenic acids in P. cocos.</p><p><b>CONCLUSION</b>The sum content of three main terpenoids in submerged cultivation mycelium P. cocos was 5. 55 times as that in cultivated material, which strongly suggested the possibility of fermentation in the production of medicinally important triterpenic acids in the future.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To offer some reference for micro-implant's development and population by analyzing clinical application of two kinds of micro-implant systems.</p><p><b>METHODS</b>38 patients treated with MIA (micro-implant anchorage) and 28 patients treated with SDIA (self-driven titanium implant for orthodontic anchorage) were included. Analyzing the rate of lost implants, the gum's reactivity and the efficiency of moving teeth summarized the excellences and shortcomings of two systems.</p><p><b>RESULTS</b>1) Six of MIA implants fell off after being inserted. Seven of SDIA implants lost when they had been implanted for a month. But they were stable after being inserted again. 2) The gum around 12 SDIA implants got inflammation symptom, but the gum around MIA implants was normal. 3) Both MIA implants and SDIA implants could move teeth effectively and persistently when they were stable.</p><p><b>CONCLUSION</b>When we apply micro-implant in clinic, we should prevent it from closing roots of teeth and choose the small tip micro-implant. The embedded position should be in area of attachment gum. At the same time, the areas around the tip of micro-implant should be keeping clean.</p>
